Learning outcome 2: Understand the kinds of influences that effect children and young people’s development. We can all be influenced by different things through life, a child’s emotional, physical, social, intellectual development can be affected by different influences, bullying, death, divorce and many more. ASSESSMENT CRITERIA 1.2 There are many different examples of how different aspects of development can have an affect on one another, here are a few examples. If a child speaks a foreign language as their first and they and their family have just moved to this country to live, this may lead to them having a language barrier, the child may be very shy, find it very hard to communicate with others and isolate themselves and finding it difficult to make friends. All of these can affect the child emotionally, socially, possibly creating behavioural problems and lowering their self-esteem, all having an effect on their development. A child who finds it difficult to interact with other children due to reasons such as bullying, may find it difficult to express how they feel, and unable to tell anyone what is happening to them. This can affect the child, emotionally they may become very upset easily or even aggressive at times, they may be affected socially, thinking that all children are going to bully them and so isolating themselves from others, and losing confidence. These may all affect their ability to concentrate in the classroom and may in turn start affecting their levels. A child who may have a disability may find that not being able to participate with certain lessons for example PE, may leave them feeling that they are being isolated and that they are different to the others, this may affect their social, behavioural and emotional development. ASSESSMENT CRITERIA 2.1 Children and young people can be affected by different kinds of influences and these can have an effect on their development. Background We all come from different backgrounds whether it be ethnic, cultural, our  environment or just circumstances. It may be that the child speaks a foreign language and is finding it difficult to communicate with others. A child may have different cultural needs to those children within the school environment. The child may come from a deprived home where there is little money and the child is not getting sufficient nutrients to sustain them during a school day. Causing lack of concentration. Also the child’s parents may be going through a divorce or separation, this can be very traumatic for a child. All of these can have an effect on the development of a child, and their ability to learn. Health Development can be affected if the child suffers with poor health or a physical disability. Low income and not being able to afford the right foods and nutrients will have an effect, physically and intellectually the child may be affected. A child with a physical disability may be emotionally and socially effected because of not being able to participate. It’s important that they are supported and included as much as possible. ASSESSMENT CRITERIA 2.2 Recognising and responding to children’s and young people’s development is important, it ensures the child or young person receives the right help and support. As soon as it is recognised that a child has a problem then the right action can be taken to ensure the child gets any help or support with their learning and they can also be monitored. If a child was ignored or the problem was not detected then their development may suffer, causing them to fall behind, or not reach their goals. You may have started to notice that a particular child is quieter than normal and isn’t their normal happy self, this could be a cause of concern, you may feel that you are able to ask the child if there is anything bothering them. They may respond by telling you that their mum and dad are not getting along and dad has left the home. This may affect the emotional behaviour of the child as well as their social as they feel they can’t talk to any of their friends about it. You may feel it’s necessary to speak to their teacher, but it’s important that this is kept as confidential. It would then be necessary to monitor the child’s development and behaviour. Being able to recognise and see a concern with a child can make all the difference to their development in the future. Being able to listen, understand and help is an important role to play as a TA.

Bacterial Transformation Using pGLO Involving X and Y Genes

Genetic transformation is due to a direct cause in the change by genes, due to the cell in taking and expressing traits from a separate piece of DNA. Naturally proficient bacteria are able to absorb exogenous DNA and go through genetic transformation. (Chen & Dubnau, 2004) The purpose of this experiment was to discover how a gene could be moved from one organism to a different organism with the help of plasmid. The cells that are capable of acquiring these traits from the other organism are known as being competent.Weedman, 2013). In this particular experiment we will genetically transform the bacteria E. coli by inserting a gene through heat shock, this gene codes for Green Fluorescent Protein, also known at GFP. The GFP gene originally comes from a Jellyfish and under an ultraviolet light the bacteria that acquired the gene with glow a brilliant fluorescent green color. (Portman et al. 2013). If the cells' nutrient medium has the sugar arabinose added to it then GFP can be turned o n. (Weedman, 2013). To determine if our hypothesis was correct, we used four differently prepared plates.The four plates each contained a different combination of the following; arabinose, ampicillin, LB nutrient broth, and pGLO plasmid. The combinations were; +pGLO LB/amp, +pGLO LB/amp/ara, -pGLO LB/amp, and -pGLO LB. Our hypothesis was: the plates with pGLO will have growth because they are resistant to the antibiotics involved, the plate with ampicillin and without pGLO will show no growth due to the fact that the antibiotic compromises the bacteria, and the plates that will grow will be the ones containing pGLO since they obtain the trait for glowing.Materials and Methods: All methods were obtained from (Weedman, 2013) Before beginning the experiment obtain latex gloves, two microcentrifuge tubes, a beaker filled with ice, a micropipetter, micropipetter tips, transformation solution containing calcium chloride, sterile loops, pGLO, E. coli, and four plates containing different s ubstances. To begin label the two microcentrifuge tubes +pGLO and – pGLO. Then proceed to obtain 250ul of transformation solution and put it in each one of the tubes using a different miropipetter tip each time, this solution will help enhance the permeability of the cell membranes.Then use a sterile loop to acquire single colony of E. coli to add to the tube labeled +pGLO; add this by twisting the sterile loop until the pGLO is off. Then repeat the last step for the -pGLO tube using a new sterile loop. Next add pGLO to the tube labeled +pGLO, to do this take a new sterile loop and inserted it into a vile containing the plasmid pGLO. Then twist the loop into the tube labeled +pGLO, then place both tubes into the beaker filled with ice for approximately 10 minutes. While the tubes are on ice grab the four LB (Luria Bertani broth) nutrient agar plates.Each plate should be labeled either +pGLO or – GLO; you should nave 1 LB/amp/ara plate (+pGLO), 1 LB plate (-pGLO 2 LB/am p plates (+pGLO)(-pGLO). After 10 minutes in the ice bath place the tubes in a floating rack and put them in a 420C water bath for exactly 50 seconds, giving them a heat shock. Immediately place both tubes back in the ice after the water bath for approximately 2 minutes. Once 2 minutes is up remove the tubes from the ice and put them in the rack at room temperature. Using a new tip each time, add 250ul of nutrient broth to both tubes. Then close the tubes and let them sit at room temperature for 10 minutes.After 10 minutes flick both tubes with your fingers to ix the contents, then using a fresh tip each time add 100ul of the transformation solution (+pGLO) and the control (-pGLO) to their appropriately labeled plates. Using a new sterile loop each time spread the contents around in each dish. Then tape the plates together and placed them upside-down in an incubator set at 370 C for 24 hours. Results: This experiment shows how a gene can be transferred from one organism to a differe nt organism through the help of plasmid. Traits are exchanged from one DNA stand toa different one in the bacteria E. coli.Two of the plates were a control group, hich meant there was no growth after the plates were taken out of the incubator. These two control plates were the ones containing -pGLO LB/amp and -pGLO LB. The transformation plates were the two plates containing +pGLO LB/amp and +pGLO LB/ amp/ara. These two plates showed a substantial growth in bacteria after being taken out of the incubator, one plate showing a considerably larger growth than the other and they both glowed under UV light due to the pGLO. Our results supported our hypothesis, the plates that showed growth were the plates containing +pGLO LB/amp and +pGLO LB/amp/ara. Where as the other two plates showed no growth at all, which matched our hypothesis. Michael Gregory did a previous experiment; he came to the same conclusion that our experiments' results oncluded.His experiment was identical to ours, involving the same materials and procedure. The same plates showed growth in his experiment as ours, as well as the plates that didn't show growth were the same. (Gregory, 2004). The only weakness that I could think of that would have a major effect on the results would be not using sterile equipment and causing cross contamination. Our experiments did not have any problems arise that would affect the results we obtained.

Today’s world is like an open book. We people now have information at our fingertips and that has helped us in drastically improving the way we live. This change in our world, into one huge global entity can be attributed to the communication revolution that our world underwent. From the radio to the computer, the developments in the field of electronics have intrigued me since childhood. My interests became my passion and motivated me to pursue my Bachelor degree in Electronics and Communication. My quest for hands-on experience motivated me in making a radio receiver in my third semester of Bachelors. But after finishing my B-Tech, I realized that I had mastered only the basics and that there is still a lot more to learn to truly comprehend the nuances of the subject. This led to my decision to pursue a Master degree and broaden my knowledge base. I believe that being able to pursue a Master Degree program in Hochschule Darmstadt will also be a motivation for me to further study and research in a field that has interested me for so long. My decision of applying to Hochschule Darmstadt, for Electrical Engineering and Information Technologies is based mainly on the course structure and the various research opportunities that I will encounter there. The eminent panel of experts handling the various topics and the resources of the university assures me of the studying experience that I hope to have. As part of my Bachelors project I did a MATLAB based project named â€Å"Copter Eyes† with introduction of a new Video Mosaicing technique. My project brought forth a comprehensive technique of real time color Video Mosaicing which invited the attention and funding of two major Institutions namely â€Å"Center for Engineering Research and Development (CERD)† and â€Å"Kerala State Council for Science Technology and Environment (KSCSTE)†. As part of the mini project of my Bachelors course, I have also worked on a microcontroller based device that works on various modes and functioned as ASCII transmitter and receiver, Pulse Width Modulator, IR Tester, Capacitance and Frequency Meter, Signal Generator, Voltage meter and Logic Probe . This small endeavor of mine was greatly appreciated by my lecturers and has been adopted by the Communication Laboratory of my college. My initial idea for the Bachelor project was a communication based project, but due to the lack of technical knowhow I was not able to go through with my project. When I could finally think again, I realized that my parents were in panic and I was surrounded by blood. Turned out, I fell right into some glass and as I would later find out, it had cut an important tendon in my shin. I was rushed to the ER and got surgery to fix my tendon. I asked the doctor to take some pictures of my leg right before I was sedated. After the surgery, a huge temporary cast was put on my leg and I was given crutches with which I could hobble around. Now came the 22-hours in a car with my injured leg. Luckily they weren’t as bad as I had thought they would be. Finally I was home. My friends visited me and were aghast at the size of my cast and the pictures of my leg! They thought I was just joking and wasn’t really injured. I had to spend 6 weeks in my cast. Six grueling weeks during which all I did was watch movies and tv shows on my laptop. My whole summer passed me by and I couldn’t do anything. My friends play basketball in the heat, thoroughly enjoying themselves while I watched them wistfully. Time went by slowly but finally I got my cast off only to find out that my tendon was too weak to walk on so I had to endure two weeks in a black boot. It was still summer and because my boot absorbed sunlight, my foot often smelled and had heat sores on it. I almost preferred my cast to this hell. Eventually my boot came off too. I could finally walk! I was sent to six weeks of physiotherapy where I found out that I probably wouldn’t be running or jumping for another 6 months almost. It merely fades such boundary lines for further publications and other journalists and the media need to take note of the code of ethics to not invade such privacy of an individual to such an extend again. The Issues Of Fresh Water In Singapore Environmental Sciences Essay Singapore is an island and urban city state with no rural hinterland and hence, it has been depending on Malaysia for nearly forty percent or more of its water supply. However, Singapore is not short of fresh water as it receives an average of around 2,400 mm of rainfall annually. The only constraint faced by the country is capturing and storing as much of this rainfall as possible, on limited amounts of land areas, so as to achieve self-efficiency in water issues. Recycling efforts in Singapore started in 1966 when the Jurong Industrial Water Treatment Plant was commissioned to supply industrial water to the Jurong Industrial Estate (JIE). This water comes from treated sewerage effluent and is meant for industrial use in the Jurong area as well as by several oil refineries. The Singapore government encourages industry and private enterprises to recycle water. Also, tax rebates have been provided for factories that install water-saving plants. Bottles of NEWater Singapore has also developed NEWater, a three-stage process which brings waste-water to a quality that is better than that produced by the Public Utilities Board (PUB). Treatment begins with Microfiltration to remove suspended solids, colloidal particles, bacteria and viruses. The next process is Reverse Osmosis, which involves applying of pressure for water to flow from the concentrated side to the less concentrated side through a semi-permeable membrane to remove inorganics like heavy metals, such as nitrate, chloride, sulphate etc. The third process, Ultra-violet disinfection, is a safety back-up to remove any bacteria or viruses that cannot be removed by Reserve Osmosis. Desalination is a process of removing excess salt from water (eg. seawater) to enable it for drinking purposes. It has been used to augment water for some time, but it was not adopted as a matter of policy until recent years. This method is considered successful, as this source of supply produce 12% of Singapores daily water needs. Although desalination costs under current improved technology are higher than traditional means of treatment, Singapore has decided to go ahead in the belief that new technologies will reduce the costs further. 3. Impoundments and reservoirs Singapore has several impounding reservoirs inland including Seletar, Peirce and MacRitchie etc. To increase storage capacity, seven reservoirs have been developed by damming the river mouths. It was in that context that a plan to maximise collection of surface resources by extending the present 24.3 square miles of collection grounds to 156 square miles (about 75% of the islands total area) was proposed. However, geological conditions limited the availability of groundwater, hence the plan focused on retrieval of stormwater and water recycling instead. Another reservoir which was built in the recent years was Marina Barrage, which was officially opened on 30th October 2008. Marina Barrage is a dam built across the Marina Channel. Water flows into the reservoir from some of Singapores most well-known waterways, including the Singapore River, Stamford Canal, Rochor Canal, Geylang River and the countrys longest river, Kallang River. Marina Barrage was envisioned by Prime Minister Lee Kuan Yew in 1987, after the great clean-up of the Singapore and Kallang Rivers. Building the barrage on the soft sea bed amidst changing currents and constantly moving marine traffic was a major challenge. From the air, the pump house of the barrage looks like that number 9, which signifies longevity in Chinese. This major engineering achievement is symbolic of Singapores development as a City of Gardens and Water. How Marina Barrage works? The Barrage acts as a physical barrier to separate the reservoir from the sea. The steel crest gates of the Barrage will act as a tidal barrier to keep the high tides out. Under normal conditions, the crest gates will remain in an upright position to isolate the reservoir from the sea. During heavy rains that collide with low tide, the steel crest will be lowered to release excess storm water into the sea. During heavy rains that collide with high tide, the steel crest gates remain erect, hence storm water cannot flow out to sea naturally. Instead, large drainage pumps will be operated to pump water out to sea. With the Barrage in place, the pockets of low-lying areas in the city will no longer be prone to flooding. 4. Stormwater run-off Harnessing stormwater at minimum cost requires proper land-use planning and pollution control to ensure that stormwater run-off can be efficiently collected and that pollution levels are low. Stormwater harvesting from urban land was consistent with the overall policy of land use in this land-scarce republic, in which land was seen as too valuable for its use to be restricted to a single purpose such as for housing. In the words of the Minister for National Development: If we designate more land for water catchments, there will be less land for housing and other developments. The reverse is also true. Optimising and maximising the use of land and water helped the country to overcome constraints to a certain extent. Among the various schemes, the Bedok and Lower Seletar Schemes are the largest. Water is conveyed to the storage reservoirs of Bedok and Lower Seletar. One important feature of this scheme is the emphasis on close co-ordination between several agencies, such as the Housing Board Development (HDB), the Ministry of the Environment (ENV) and the Planning Department, in order to ensure that water collected from urban surfaces is low in pollutants. In addition, the drainage system designed in such a way that it channels water through concrete channels to suitable collection ponds situated at topographic low points. An automatic monitoring system ensures that only discharges produced by heavy storms resulting in run-o ff above a certain volume are collected. One example of the innovative approach to stormwater collection is making use of the empty space under the interchange of the Seletar and Bukit Timah expressways for a water collection pond. 5. Conservation of water In addition to developing and diversifying water resources, water conservation is also key to ensuring a sustainable water supply.
